Annotation score:5 out of 5

The annotation score provides a heuristic measure of the annotation content of a UniProtKB entry or proteome. This score cannot be used as a measure of the accuracy of the annotation as we cannot define the 'correct annotation' for any given protein.

-Experimental evidence at protein leveli
This indicates the type of evidence that supports the existence of the protein. Note that the 'protein existence' evidence does not give information on the accuracy or correctness of the sequence(s) displayed.

This section provides any useful information about the protein, mostly biological knowledge.
Functioni

Cytochrome P450 monooxygenase involved in the biosynthesis of ergosterol (PubMed:18191972, PubMed:26459890, PubMed:29439966).

The existence of several duplicated sterol 14-alpha demethylase genes could be a good strategy to modulate the composition and fluidity of the cell membrane (By similarity).

Catalyzes C14-demethylation of erburicol to produce 4,4,24-trimethyl cholesta-8,24(28)-dien-3-beta-ol (PubMed:18191972, PubMed:26269599).

Shows a slight substrate preference toward eburicol over obtusifoliol (PubMed:26269599).

As a target of azole drugs, plays a crucial role in azole susceptibility (PubMed:12543662, PubMed:26269599, PubMed:28461309, PubMed:29894182).

This subsection of the 'Function' section provides information relevant to cofactors. A cofactor is any non-protein substance required for a protein to be catalytically active. Some cofactors are inorganic, such as the metal atoms zinc, iron, and copper in various oxidation states. Others, such as most vitamins, are organic.
Cofactori

The activity is completely inhibited by miconazole and reduced by voriconazole (6% substrate conversion), posaconazole (10%), itraconazole (16%), (R)-N-(1-(2,4-dichlorophenyl)-2-(1H-imidazol-1-yl)ethyl)-4-(5-phenyl-1,3,4-oxadiazol-2-yl)benzamide (VNI) (20%), and (R)-N-(1-(3,4'-difluorobiphenyl-4-yl)-2-(1H-imidazol-1-yl)ethyl)-4-(5-phenyl-1,3,4-oxadiazol-2-yl)benzamide (VFV) (31%). The weakest inhibitory effect is observed with fluconazole (PubMed:26459890, PubMed:26269599, PubMed:28461309). Activity is also inhibited by ketoconazole, as well as by the azole-based drug candidates VT-1161 and VT-1598 (PubMed:28461309). Activity is inhibited by the novel and long-acting fungicidal azole PC1244 (PubMed:29439966).4 Publications

The checksum is a form of redundancy check that is calculated from the sequence. It is useful for tracking sequence updates.

It should be noted that while, in theory, two different sequences could have the same checksum value, the likelihood that this would happen is extremely low.

However UniProtKB may contain entries with identical sequences in case of multiple genes (paralogs).

The checksum is computed as the sequence 64-bit Cyclic Redundancy Check value (CRC64) using the generator polynomial: x64 + x4 + x3 + x + 1.
