Functioni

Belongs to the chromatin remodeling brain-specific BAF (bBAF) complex, as such plays a role in remodeling mononucleosomes in an ATP-dependent fashion. Belongs to the neuron-specific chromatin remodeling complex (nBAF complex) and is required for postmitotic neural development and dendritic outgrowth. During neural development a switch from a stem/progenitor to a post-mitotic chromatin remodeling mechanism occurs as neurons exit the cell cycle and become committed to their adult state. The transition from proliferating neural stem/progenitor cells to post-mitotic neurons requires a switch in subunit composition of the npBAF and nBAF complexes. As neural progenitors exit mitosis and differentiate into neurons, npBAF complexes which contain ACTL6A/BAF53A and PHF10/BAF45A, are exchanged for homologous alternative ACTL6B/BAF53B and DPF1/BAF45B or DPF3/BAF45C subunits in neuron-specific complexes (nBAF). The npBAF complex is essential for the self-renewal/proliferative capacity of the multipotent neural stem cells. The nBAF complex along with CREST plays a role regulating the activity of genes essential for dendrite growth. ACTL6B/BAF53B is not essential for assembly of the nBAF complex but is required for targeting the complex and CREST to the promoter of genes essential for dendritic growth.3 Publications

Tissue specificityi

Restricted to the brain and peripheral nervous tissue (at protein level). Present in virtually all neurons in the cerebral neocortex (layers II-VI), hippocampus (CA1-CA3 region and dentate gyrus), cerebellum (molecular, granular and Purkinje cell layers), spinal cord (dorsally and ventrally), dorsal root ganglion, retina and the olfactory bulb (mitral and granule cell layers). Expressed specifically in post-mitotic neurons (at protein level).3 Publications

Developmental stagei

Expression begins near the time of neuronal cell type specification. First apparent at 10.5 dpc in the nervous system, with high levels in the telencephalon and less in the diencephalon, mesencephalon and spinal cord. This pattern of expression persists and becomes more defined at 12.5 dpc and later stages. Detected in dorsal root ganglia, but not in neural crest derivatives that give rise to non-neuronal tissues, including great vessels. Expressed in postmitotic cells in the CNS, but not in actively proliferating precursor cells. At protein level, first detected at 12.5 dpc in nervous tissues. In the developing forebrain, cerebellar primordium and spinal cord, strictly expressed in post-mitotic neurons.2 Publications
