General annotation (Comments)

Function

Calcium/calmodulin-dependent protein kinase belonging to a proposed calcium-triggered signaling cascade. In vitro phosphorylates transcription factor CREB1.

Catalytic activity

ATP + a protein = ADP + a phosphoprotein. Ref.1

Enzyme regulation

Activated by Ca2+/calmodulin. Binding of calmodulin is thought to result in a conformational change and leads to activation through phosphorylation by CAMKK1. Ref.1

Subcellular location

Cytoplasm. Golgi apparatus membrane; Peripheral membrane protein. Cell membrane; Peripheral membrane protein Ref.1.

Tissue specificity

Highly expressed in brain, in neuronal cell bodies of the central nucleus of amygdala and ventromedial hypothalamic nucleus. Also detected in heart, testis, and kidney. Ref.1

Developmental stage

Expression starts at E11 in parallel with the onset of development of the central nervous system. Ref.1

Domain

The autoinhibitory domain overlaps with the calmodulin binding region and interacts in the inactive folded state with the catalytic domain as a pseudosubstrate By similarity.

Post-translational modification

May be prenylated on Cys-474.

Sequence similarities

Belongs to the protein kinase superfamily. CAMK Ser/Thr protein kinase family. CaMK subfamily.

Contains 1 protein kinase domain.
