General annotation (Comments)

Function

Poly [ADP-ribose] polymerase using NAD+ as a substrate to transfer ADP-ribose onto glutamic acid residues of a protein acceptor; repeated rounds of ADP-ribosylation leads to the formation of poly(ADPribose) chains on the protein, thereby altering the function of the target protein. May play a role in the adaptative response to chemical exposure (TCDD) and thereby mediates certain effects of the chemicals. Ref.1

NAD+ + (ADP-D-ribosyl)(n)-acceptor = nicotinamide + (ADP-D-ribosyl)(n+1)-acceptor.

Induction

By 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD), which is a by-product of industrial processes and an important environmental contaminant. This induction is concentration and time-dependent and is mediated through an aryl hydrocarbon receptor (AhR) and aryl hydrocarbon receptor nuclear translocation (Arnt) signal transduction. Superinduced by cycloheximide and by inhibitors of the 26S proteasome. Ref.1 Ref.4
