General annotation (Comments)

Function

Inhibitor of the E3 ubiquitin-protein ligase ITCH. Acts by interacting with the second WW domain of ITCH, leading to compete with ITCH's substrates and impairing ubiquitination of substrates By similarity.

Subunit structure

Interacts with NEDD4. Interacts with ITCH (via WW domain 2) By similarity.

Subcellular location

Nucleusnucleolus By similarity. NucleusPML body By similarity. Note: Primarily localizes to the nucleolus. Also localizes to the PML nuclear bodies, when desumoylated By similarity.

Tissue specificity

Detected in heart, lung, brain, liver, skeletal muscle, pancreas, kidney, spleen, testis and ovary. Ref.1

Post-translational modification

Monoubiquitinated by NEDD4. Polyubiquitinated, leading to its degradation by the proteasome. Sumoylated by SUMO1, abrogating polyubiquitination and subsequent degradation. Desumoylated by SENP1, leading to accumulation in PML nuclear bodies By similarity.

Phosphorylated upon DNA damage, probably by ATM or ATR. Ref.4 Ref.5 Ref.6 Ref.7

Sequence similarities

Belongs to the N4BP1 family.

Sequence caution

The sequence BAA31590.2 differs from that shown. Reason: Erroneous initiation. Translation N-terminally shortened.
