General annotation (Comments)

Function

ATP dependent phosphorylation of adenosine to monophosphate derivatives. The enzyme exculusively uses adenosine or methyl-adenosine, and prefers GTP and dGTP to ATP. Ref.2

Catalytic activity

ATP + adenosine = ADP + AMP.

Cofactor

Magnesium. Ref.2

Enzyme regulation

The enzyme is subject to inhibition by adenosine and is competitively inhibited by the adenosine analog idotubercidin. Unlike other adenosine kinases it is not stimulated by inorganic phosphate. Ref.2

Activity is stimulated in the presence of potassium, resulting in a 13-fold increase in the catalytic efficiency for adenosine.

KM=79 µM for methyl-adenosine (in the presence of 10 mM KCl)

KM=1100 µM for ATP (in the presence of 10 mM KCl)

Vmax=180 nmol/min/mg enzyme (for adenosine in the presence of 10 mM KCl)

Vmax=72 nmol/min/mg enzyme (for methyl-adenosine in the presence of 10 mM KCl)

pH dependence:

Optimum pH is 8-11.
